چکیده

There are two major cancer susceptibility genes, BRCA1 and BRCA2, mutations in which predispose to early onset breast and ovarian cancer. 2 The frequency of BRCA1 and BRCA2 mutations in the general population ranges from 1-500 to 1-800. 3 In a recent population study, the authors showed that the estimated population frequencies for BRCA1 and BRCA2 mutations were similar under both recessive and polygenic models, 0.024 and 0.041%, respectively. These frequencies are lower than the frequencies found before. 3 Therefore, the prior probability of finding any person or family with mutations in both the BRCA1 and BRCA2 genes is very low. In the Ashkenazi Jewish population, however, the likelihood of being a carrier of one of the three common BRCA1 or BRCA2 mutations is as high as 1 in 50; this explains why there are multiple reports of double heterozygotes for these mutations. There have been reports of families with two mutations, subjects with two mutations, and families with three mutations in the BRCA genes, mostly in the Ashkenazi Jewish population. To date, only a few subjects or families have been reported to have more than one non-Ashkenazi BRCA mutation. There are five reports describing families that harbour two BRCA1 or one BRCA1 and one BRCA2 mutation. 12–14 We report the first Spanish breast cancer family where two independent mutations, one in BRCA1 and the second in BRCA2, are present in multiple members of a single sibship. The two mutations were found in many subjects. Analysis of the pedigree showed a spectrum of cancer phenotypes associated with one or two mutations, as well as different ages of onset of the cancer.

The importance of BRCA1 and BRCA2 genes mutations in breast cancer development

Many factors including genetic, environmental, and acquired are involved in breast cancer development across various societies. Among all of these factors in families with a history of breast cancer throughout several generations, genetics, like predisposing genes to develop this disease, should be considered more. Early detection of mutation carriers in these genes, in turn, can play an import...

Minor role of BRCA2 mutation (Exon2 and Exon11) in patients with early-onset breast cancer amongst Iranian Azeri-Turkish women

Objective(s): Breast cancer is the most common cancer in women. Every year, one million new cases are reported worldwide, representing 18% of the total number of cancer in women. Hereditary BRCA1 and BRCA2 mutations account for about 60% of inherited breast cancer and are the only known causes of hereditary breast cancer syndrome. The aim of this study was to determine the frequency of BRCA2 (e...

بررسی موتاسیون های شایع ژن BRCA1 و BRCA2 در دختران مادران مبتلا به سرطان سینه و مقایسه آن با گروه کنترل

Background and purpose: Breast Cancer is one of the health problems in every population. The aim of this study was to determine the frequency of BRCA1 and BRCA2 common mutations in women whose mothers were diagnosed with breast cancer. Materials and methods: A case–control study was performed in 109 females (less than 40 years of age) who had mothers with breast cancer. For scanning of...

جهش های ژنتیکی جدید در ژن های اصلی سرطان پستان (BRCA1/BRCA2) در زنان ایرانی مبتلا به سرطان پستان زودرس

Background: Breast cancer is the most common female malignancy and the main cause of death in  mid-aged women. Genetic germ line mutations in BRCAI/BRCA2 in Iranian women with breast or  ovarian cancer have not been yet reported. Materials and methods: Clinical data, family history and blood samples were obtained from 83  females aged less than 45 years with primary breast cancer in order to su...

Women with hereditary breast cancer predispositions should avoid using their smartphones, tablets and laptops at night

Breast cancer is the most common malignancy among women, both in the developed and developing countries. Women with mutations in the BRCA1 and BRCA2 genes have an increased risk of breast and ovarian cancers. Recent studies show that short-wavelength visible light disturb the secretion of melatonin and causes circadian rhythm disruption. We have previously studied the health effects of exposure...
